Kind of depends on the tone and direction of said reboot or sequel. Smurfette and Brainy always seem to be included, but the other characters can vary a bit.
The good qualities don’t get displayed much. Usually I’m entirely sympathetic with whichever Smurf has tossed Brainy out on his head or tail.

@RedPixel64DS you talking about where the 80s cartoon left off? I doubt that would get a follow up movie. It’d require a ton of explanation for folks not familiar with that season. And people who are familiar with the season probably wouldn’t want a follow up for the most part. Paul Winchell, original voice of Gargamel, left the show over how bad it was.
I thought The Lost Village was good, myself.
At any rate, I’d probably take its approach over what the other movies did/are going to do. No trips to modern times, no blend of live action and animation.
So I was watching “The Pussywillow Pixies” again, and it struck me that you could work the Smurf Girls into a new version of that story quite nicely. The Smurf Girls’ penchant for dressing up in masks and their general fierceness could easily explain the reputations Smurfs have among the pixies and Wartmongers.
I could go for Balthazar returning.
So I was watching The Astrosmurf when this conversation came up:

@The NickToon Dude 100 Well, judging by its Rotten Tomatos page, not a lot of people were impressed by the second live-action film: The Smurfs 2 - Rotten Tomatoes Personally, I don't mind, as I didn't like the looks of either live-action film. The Lost Village is a vast improvement in my opinion, and while it apparently didn't do well enough critically to receive a sequel either, it did at least introduce some characters that the Smurfs franchise is apparently intent on keeping around.
Huh...an interesting sample. Can't say I'm sorry to see it not get made, though, if it was going to be part of the live-action/computer animation hybrid movie series. I'll take Smurfwillow over a new female Smurf from that version of the Smurfs anyday.
Do we have any confirmation that the saxophone player in The Lost Village is supposed to be Smooth Smurf? Personally I wonder if it isn't meant to be Harmony, having found his true instrument at long last.

I don’t get what a prequel would be about. They’re usually origin stories, but it’s not like the Smurfs have an origin. There’s Smurfette, I guess, but hers has kind of been done to death.
Then again, if it were set in The Lost Village/2021 series universe, I suppose they could do a story about how the Smurfs and Smurfettes came to be living in separate villages or something; maybe reintroduce Grandpa and Nanny while they’re at it.
@Jbarth I get the idea. Though, from the sounds of things, it seems more likely that the Pluffs would end up manipulating Gargamel as opposed to being recruited as his "army".
@Jbarth nice Lion King reference. I hardly think Gargamel is the equal of Scar.
